Physiological and cellular responses in two populations of the mussel Perna perna collected at different sites from the coast of São Paulo, Brazil BABT
Abessa,Denis Moledo de Souza; Zaroni,Letícia Pires; Sousa,Eduinetty Ceci Pereira Moreira de; Gasparro,Marcia Regina; Pereira,Camilo Dias Seabra; Rachid,Bauer Rodarte de Figueredo; Depledge,Michael; King,Rebecca Susan.
The physiological conditions of mussels from Ubatuba and Santos and also of organisms transplanted from Ubatuba to Santos were studied by using different techniques. Assays for lysosomal stability were conducted on the haemolymph. Heart rate activity was monitored for 6h. The embryonic development of larvae obtained from the collected mussels was analysed. For all the compared groups of mussels, no significant differences were observed for the cardiac activity monitoring and the embryonic bioassays. The mean Neutral Red (NR) retention time was similar for the animals from Santos and Ubatuba, whereas the organisms transplanted to Santos showed a reduction in the retention time of the dye, indicating damage in the lysosomal membranes. These differences were...

Biomarkers for screening of lung cancer and pre-neoplastic lesions in a high risk Chilean population Biol. Res.
Adonis,Marta I; Díaz,Jose; Miranda,Veronica R; Chahuan,Marco; Zambrano,Alcides; Benitez,Hugo C; Campos,Monica; Avaria,Pablo; Urzúa,Ulises; Marín,Pedro; Gohurdett,Mariela; Cisterna,Yasna; Gil,Lionel.
BACKGROUND: The mortality of lung cancer (LC), increases each year in the world, in spite of any advances, in development of new drugs to advance stages of LC. The high incidence of LC has been associated with smoking habit, genetic diversity and environmental pollution. Antofagasta region has been reported to have the highest LC mortality rate in Chile and its inhabitants were exposed to arsenic in their drinking water in concentrations as high as 870 μg/L. Non-invasive techniques such as biomarkers (Automatic Quantitative Cytometry: AQC and DR70) and Auto Fluorescence Bronchoscopy (AFB) might be potentially useful as a supplementary diagnostic approach and early detection. Early detection is one of the most important factors to intervene and...

Akcha, Farida.
In the aim to develop biomarkers of genotoxicity for the biomonitoring of the marine environment, the formation of DNA adducts and oxidized DNA bases was studied for the sentinel organism, Mytilus sp, following exposure to B[a]P via the trophic supply. DNA adducts and 8-oxo-7,8-dihydro-2'-désoxyguanosine (8-oxodGuo) were respectively measured by 32P post-labeling technique and high performance liquid chromatography coupled to electrochemical detection. The developped model of exposure was validated for the realisation of toxicological studies in Mytilus sp. The bioavailability of B[a]P for the exposed organisms and its biotransformation resulted in variations in most of the measured enzymatic biomarkers of exposure. Such an exposure allowed the expression...

Trends in biological dosimetry: an overview BABT
Amaral,Ademir.
Biological dosimetry (biodosimetry) is based on investigations of induced biological effects (biomarkers) in order to correlate them with radiation dose. Among the indicators employed in biodosimetry, scoring of chromosome aberrations is the most reliable method to quantify individual exposure to ionizing radiation. The technique, applied to circulating lymphocytes, has been developed into a routine procedure to evaluate the dose in the case of real or suspected accidental exposure. Considering the radiosensitivity of lymphocytes in vitro and in vivo as being the same, the dose effect relationship obtained after in vitro irradiation of blood has been widely used, with medico-legal value, for evaluating individual radiation exposure. This report presents an...

Distribution of the organic matter in the channel-levees systems of the Congo mud-rich deep-sea fan (West Africa). Implication for deep offshore petroleum source rocks and global carbon cycle ArchiMer
Baudin, Francois; Disnar, Jean-robert; Martinez, Philippe; Dennielou, Bernard.
The quantity and the source of organic matter preserved in the Recent turbiditic channel-levees systems around 4000 m-depth off the Congo River were determined using bulk geochemical approaches (Rock-Eval, elemental and isotopic analyses) as well as molecular and optical analyses on selected samples. These mud-rich sediments contain high amount of organic matter (3% Corg on average), the origin of which is a mixture of terrestrial higher-plant debris and deeply oxidized phytoplanktonic material. Although the relative contribution of continental source versus marine source of the organic matter cannot be precisely quantified, the continental fraction appears significant (at least 70-80%) especially for such depths and distances from the coast. The organic...

Fatty acid composition in deep hydrothermal vent symbiotic bivalves. ArchiMer
Ben-mlih, F; Marty, Jc; Fiala-médioni, A.
Fatty acids in deep hydrothermal vent bivalves have been analyzed. Their composition is completely different from that of a littoral mussel collected in the Mediterranean sea. The distribution of fatty acids in the littoral mussel is characterized by a predominance of polyunsaturated fatty acids (20:5n-3, 22:6n-3) reflecting the planktonic origin of the food. Vent bivalve fatty acid distribution is dominated by an abundance of the monounsaturated acids (double bond in the n-7 position) 16:1n-7, 18:1n-7, and 20:1n-7 which are clearly of bacterial origin and give an indication of the symbiotic bacterial activity in the bivalves. Differences between the fatty acid composition of the bivalves from two hydrothermal sites (13 degrees N and Galapagos) and...

Fatty acids from lipids of marine organisms: Molecular biodiversity, roles as biomarkers, biologically active compounds, and economical aspects ArchiMer
Berge, Jean-pascal; Barnathan, Gilles.
Because of their characteristic living environments, marine organisms produce a variety of lipids. Fatty acids constitute the essential part of triglycerides and wax esters, which are the major components of fats and oils. Nevertheless, phospholipids and glycolipids have considerable importance and will be taken into account, especially the latter compounds that excite increasing interest regarding their promising biological activities. Thus, in addition to the major polyunsaturated fatty acids (PUFA) such as eicosapentaenoic (EPA) and docosahexaenoic (DHA) acids, a great number of various fatty acids occur in marine organisms, e.g. saturated, mono- and diunsaturated, branched, halogenated, hydroxylated, methoxylated, non-methylene-interrupted. Various...

Cultured Pearl Surface Quality Profiling by the Shell Matrix Protein Gene Expression in the Biomineralised Pearl Sac Tissue of Pinctada margaritifera ArchiMer
Blay, Carole; Planes, Serge; Ky, Chin-long.
Nucleated pearls are produced by molluscs of the Pinctada genus through the biomineralisation activity of the pearl sac tissue within the recipient oyster. The pearl sac originates from graft tissue taken from the donor oyster mantle and its functioning is crucial in determining key factors that impact pearl quality surface characteristics. The specific role of related gene regulation during gem biogenesis was unknown, so we analysed the expression profiles of eight genes encoding nacreous (PIF, MSI60, PERL1) or prismatic (SHEM5, PRISM, ASP, SHEM9) shell matrix proteins or both (CALC1) in the pearl sac (N = 211) of Pinctada margaritifera during pearl biogenesis. The pearls and pearl sacs analysed were from a uniform experimental graft with sequential...

Biological effects of the "Erika" oil spill on the common mussel (Mytilus edulis) ArchiMer
Bocquene, Gilles; Chantereau, Sebastien; Clerendeau, Christelle; Beausir, Emilie; Menard, Dominique; Raffin, Bernard; Minier, Christophe; Burgeot, Thierry; Leszkowicz, Annie Pfohl; Narbonne, Jean-françois.
A 3-year survey was made of several biological markers in mussels (Mytilus edulis) exposed in situ to the oil that came ashore after the wreck of the "Erika" tanker on the Brittany (France) coast in December 1999. The mussel response was assessed using a set of 7 biomarkers, most of them related to the metabolism of organic contaminants. After a series of validation tests, data was evaluated for only 5 biomarkers: acety1cholinesterase (AChE), glutathione S-transferase (GST), catalase (CAT), malondialdehyde (MDA) and deoxyribonucleic acid (DNA) adducts. No significant reductions in GST or CAT levels were observed, levels of DNA adducts and MDA were high during the 6 months immediately following the accident and levels of AChE were significantly lower during...

Etude des variations saisonnières de différents biomarqueurs, chez la moule Mytilus galloprovincialis. Relation avec certains paramètres physiques, chimiques et physiologiques ArchiMer
Bodin, Nathalie.
The aim of this study was to evaluate the seasonal variations of six biomarkers on Mytilus galloprovincialis, using comparison with physiological index and chemical contamination levels on two sites of  Mediterranean sea. The seasonal variations of acetylcholinesterase (AChE), DNA adducts, benzo(a)pyrene hydroxylase (BPH), heat-shock proteins (HSP), metallothioneins (MT) and multixeniobiotic proteins (MXR) were estimated over a two-year period, on mussels which were sampled in Carteau (original site) and La Fourcade (transplanted organisms). Carteau, an industrial site, presents higher contamination levels (HAP, PCB, heavy metals) than La Fourcade, an urban area. The PAH profiles underline a strong spatio-temporal variability of levels and sources of...

Association between soluble biomarkers - microbial translocation, inflammation and cardiovascular risk in HIV- infected individuals: a systematic review BJPS
Borato,Danielle Cristyane Kalva; Vellosa,José Carlos Rebuglio.
Microbial translocation is associated with the increased risk of cardiovascular disease in HIV-infected individuals. There is scarce information regarding the possible associations between the biomarkers of microbial translocation, inflammation and cardiovascular risk that can be evaluated in clinical laboratories using plasma or serum samples. This systematic review was conducted according to the PRISMA protocol in order to verify the most used soluble biomarkers of microbial translocation, inflammation and cardiovascular risk, as well as possible associations between them, in HIV-infected individuals. A search was performed using the Medline, Scopus and Web of Science databases to identify existing studies regarding the relationship between microbial...

The objective of this study was to develop an integrative approach in ecotoxicology (from biomarkers to population genetics) to assess the ecological status of fish populations. Flounders (Platichthys flesus) collected after the spawning season in the heavily polluted Seine estuary were compared with the moderately polluted Bay of Douarnenez. The muscle energetic reserves were highly depleted in Seine vs. Douarnenez fish. The Seine fish displaying a reduced capacity to manage the oxidative stress and a higher energetic metabolism. An increase in the content of muscle membrane phospholipids (sphingomyelin, phosphatidylserine, free sterols) was detected in the Seine vs. Douarnenez fish. The data integration allowed to hypothesize relationships between...

Engineered nano-sized Cu oxide particles are extensively used in diverse applications. Because aquatic environments are the ultimate "sink" for all contaminants, it is expected that nanoparticles (NP) will follow the same fate. In this study, two marine invertebrates Scrobicularia plana and Hediste diversicolor were chosen as ecotoxicological models. The aim was to evaluate behavioural (burrowing kinetics, feeding rate) and biochemical (biomarkers) responses of S. plana and H. diversicolor exposed in the laboratory to Cu (10 mu g L(-1)) added in natural seawater either in the form of engineered nanoparticles (NPs) of CuO or as dissolved Cu in 2% HNO(3). Exposure was characterized by considering (i) the physico-chemical fate of NP (ii) the fraction of...

In the Atlantic French coast, significant mortalities were observed in oyster communities during gametogenesis. These deaths possibly result from the cumulative effects of various stressors such as seasonal temperature variations, pesticide application and the release of contaminants from the resuspension of sediments. A field survey, based on aquaculture conditions at 15 and 70 cm from the sediment bed with genetically-resistant (R) and -sensitive (S) lines, showed a stress condition. Significant weight loss was observed during May 14th through June 10th for both R and S lines. On one hand, the levels of the egg yolk vitellin in females were decreased at May 14th for both R and S lines. On the other hand, a significant increase in vitellin production was...

Detection of multidrug-resistant Mycobacterium tuberculosis strains isolated in Brazil using a multimarker genetic assay for katG and rpoB genes BJID
Café Oliveira,Luita Nice; Muniz-Sobrinho,Jairo da Silva; Viana-Magno,Luiz Alexandre; Oliveira Melo,Sônia Cristina; Macho,Antonio; Rios-Santos,Fabrício.
Abstract Multidrug-resistant tuberculosis (MDRTB) is a serious world health problem that limits public actions to control tuberculosis, because the most used anti-tuberculosis first-line drugs fail to stop mycobacterium spread. Consequently, a quick detection through molecular diagnosis is essential to reduce morbidity and medical costs. Despite the availability of several molecular-based commercial-kits to diagnose multidrug-resistant tuberculosis, their diagnostic value might diverge worldwide since Mycobacterium tuberculosis genetic variability differs according to geographic location. Here, we studied the predictive value of four common mycobacterial mutations in strains isolated from endemic areas of Brazil. Mutations were found at the frequency of...
